浸矿微生物在黄铁矿氧化中的“接触作用”研究进展

摘 要:
黄铁矿是地球表面最丰富的硫化矿, 它的氧化与有色金属提取工业和酸性水治理密切相关。一方面, 对于难处理金矿石, 金矿物浸染状分布于黄铁矿中, 有效地氧化黄铁矿是金矿物释放的关键。另一方面, 生物堆浸体系中黄铁矿的氧化会不断产酸, 增加中和成本。浸矿微生物既存在于矿物表面又存在于溶液中, 对于黄铁矿的氧化发挥着各自的作用。浸矿微生物在黄铁矿表面的吸附/脱附, 表面胞外聚合物 (EPS) 的形成以及定殖是生物氧化黄铁矿的关键环节, 同时其机制也是促进或抑制工业体系中黄铁矿氧化的理论依据。然而微生物与黄铁矿表面作用力的实质, 胞外聚合物 (EPS) 组成的调控机制, 以及这些吸附于黄铁矿表面的浸矿微生物对于黄铁矿生物氧化中所扮演的角色还不甚明了。本文综合国内外文献报道, 着重讨论浸矿微生物与黄铁矿的“接触作用”中各个环节及其所扮演角色, 根据目前研究现状总结了存疑问题并提出了未来研究方向。

Research Progress in “Contact Mechanism” of Bioleaching Bacteria on Pyrite Oxidation

Abstract:
Pyrite is the most abundant metal sulfides in the world. Its oxidation is related to the economic benefit of extractive metallurgy of nonferrous metal and the control of acid mine drainage. On the one hand, effective oxidation of pyrite is a key factor to the release of gold minerals from refractory gold ores, for which gold minerals disseminate in the pyrite matrix. On the other hand, oxidation of pyrite in bioheap leads to the generation of acid, which increases the neutralization cost. Sessile and planktonic bacteria play their respective role in pyrite oxidation. The attachment/detachment, formation of extracellular polymeric substance ( EPS) and colonization of bioleaching bacteria on pyrite are key segments of pyrite bioxidation and its mechanism is the basis of commercial operation to decide either promoting or inhibiting the oxidation of pyrite. However, the essence of force between bacteria and pyrite surface, the regulatory mechanism of EPS composition and the detail role of the bacteria that attach to the pyrite surface are still unknown. Combining domestic and foreign literatures, the review focused on the processes of “contact mechanism”and the role of “contact bacteria”. Controversial issues were summarized, and the new direction in the future was proposed.

黄铁矿常伴生于黄铜矿、辉铜矿、闪锌矿等硫化矿以及稀有金属金等。它的氧化与工业利润和环境问题直接关联。一方面, 对于浸染状分布于黄铁矿中的金矿物, 黄铁矿的有效氧化是金释放的关键。另一方面, 生物堆浸体系中黄铁矿的氧化会不断产酸, 增加中和成本。此外, 黄铁矿的氧化是酸性水的主要来源, 此时抑制黄铁矿的氧化是控制酸性水产生的关键。
黄铁矿的氧化与其共存体系中的微生物关系密切, 而微生物在黄铁矿氧化中所扮演的角色一直是一个具有争议的议题。最初Ehrlich和Silverman
[1 ]
提出直接/间接作用假说解释微生物在浸出中的作用。而随着研究的深入, 不同学者从不同的角度阐明微生物酶系统直接氧化硫化矿中硫为基础的“直接作用”并不存在
[2 ,3 ,4 ,5 ,6 ]
。Sand等
[7 ,8 ]
学者在分析硫化矿物分子轨道与价带特征, 硫化矿氧化过程中间态硫组分以及胞外聚合物 (EPS) 组成的基础上把之前微生物的间接作用划分为接触/非接触作用。目前对于微生物的非接触作用已被广大学者所认可, 而接触作用还存在争议。微生物的接触作用对于黄铁矿的氧化以及整个生物堆浸体系设计思路有至关重要的影响, 对其机制的研究是目前生物冶金中研究热点之一

。本文针对微生物/黄铁矿“接触作用”这一科学问题, 将近年来国内外报道的该领域有关界面作用的研究成果作一综述, 以作参考。
1浸矿微生物在黄铁矿溶解中的作用
黄铁矿是一种重要且分布最广的硫化矿, 而微生物在硫化矿溶解中的作用一直是有争议的议题。在过去40多年中, 关于微生物通过“直接/间接”作用氧化硫化矿存在持续争议。Rohwerder和Sand
[2 ]
认为Schippers等
[3 ,4 ,5 ,6 ]
对硫化矿氧化背后的硫化学转化机制的阐明结束了近几十年来关于微生物在硫化矿氧化中的“直接/间接”作用的争论。Sand和他的同事
[7 ,8 ]
认为硫化矿氧化过程中微生物只存在“间接”作用, 即Fe3+ 是氧化硫化矿的氧化剂而微生物则是将体系中的Fe2+ 和硫化矿溶解的中间态硫化产物分别氧化为Fe3+ 和硫酸根, 而不存在“直接”作用, 即微生物直接通过酶系统氧化硫化矿中的硫。基于硫化学的理论分析以及微生物胞外聚合物的组成分析, 他们将微生物的“间接”作用进一步划分为“接触/非接触”作用。
近年越来越多的学者将生物浸出机制归为接触/非接触/联合作用浸出机制
[10 ,11 ,12 ,13 ,14 ]
。黄铁矿是酸不可溶硫化矿
[5 ]
, 酸性体系下溶液氧化还原电位是影响其氧化速率的最重要因素
[15 ,16 ,17 ]
, 其氧化机制满足电化学反应控制机制
[13 ,14 ,18 ,19 ,20 ]
。而微生物在黄铁矿的溶解中的非接触作用是Fe3+ 与黄铁矿反应生成二价铁和多种形式的硫化物, 游离于溶液中的微生物可以以Fe2+ 作为电子给予体, O2 为电子受体, 氧化铁再生高铁, 式 (1) 所示, 从而提高溶液中的氧化还原电位继续氧化黄铁矿, 如式 (2) 。
接触作用为吸附于硫化矿物表面的微生物以胞外聚合物 (EPS) 为反应空间进行的高铁再生从而继续氧化矿物的过程。矿物溶解过程发生于细菌与矿物之间的EPS内, 此作用的存在证据主要为两个方面, 其一是矿物局部产生细菌形状的腐蚀坑
[12 ,21 ,22 ]
, 第二是提取出的EPS中存在三价铁复合物。然而对于微生物的“接触作用”还有存疑。目前的研究局限于未能完全避免“非接触作用”的影响, 即矿物表面的腐蚀坑的形成可能源于Fe3+ 离子的作用, 而EPS的提取环境中也存在大量Fe3+ 离子。Fowler等
[18 ,23 ]
使用电化学方法控制溶液本体电位和p H值比较有菌/无菌体系下黄铁矿氧化速率, 发现有菌体系快于无菌, 并将其归因于吸附于黄铁矿表面的细菌可以提高矿物表面的p H值。联合作用机制是指
[24 ]
对于酸不可溶硫化矿 (黄铁矿) , 一些吸附于表面生物膜中的细菌联合溶液中游离态的细菌同时发挥作用再生高铁, 从而氧化酸不溶硫化矿, 即接触/非接触作用同时存在。
目前对于接触作用可能的机制主要有两种推测
[25 ]
。一种推测观点认为由于矿物和EPS的反应界面产生的二价铁在EPS中形成的二价铁葡萄醛酸络合物不稳定, 一部分二价铁葡萄醛酸络物分解释放Fe2+ 至EPS之外, 剩余的葡萄糖醛酸复合物残基结合附近溶液中 (EPS外) 的三价铁形成较二价铁络合物稳定的三价铁络合物;另一部分产生的二价铁在EPS中扩散至细菌外膜重新被氧化为三价铁形成络合物。另一种观点认为EPS中的三价铁通过电子隧穿效应
[26 ]
得到矿物表面2 nm之内的电子从而被还原。然而人们目前对于接触作用的原子级机制还知之甚少。
2微生物吸附及其与矿物相互作用
不同的学者对于浸矿微生物与矿物的相互作用过程有不同具体细节的描述。而浸矿微生物与矿物相互作用大致可以分为4个不同的阶段: (1) 细菌扩散至矿物表面, (2) 起始的吸附, (3) 牢固的吸附, (4) 表面定殖
[27 ]
。而微生物与矿物的作用类型又可以分为物理/化学作用, 如静电力作用, 亲/疏水作用和生物作用, 如化学感应系统。
在第一阶段, 微生物通过扩散, 对流, 沉降, 细胞运动等方式接近矿物表面。在接近矿物表面时, 不同类型的界面力控制吸附过程。大于50 nm时, 细菌与基质只有范德华力;10~20 nm时, 细菌与基质之间既有范德华力又有静电力的相互作用;当细菌与基质的距离接近甚至小于1.5 nm时, 除了范德华力与静电力之外, 细菌分泌的胞外聚合物 (EPS) 中的大分子基团会与基底发生特殊相互作用, 例如形成疏水键等。第二阶段为起始吸附阶段, 起始阶段的吸附可以分为可逆和不可逆吸附。Gehrke等
[28 ,29 ]
和Kinzler等
[30 ]
研究了吸附于黄铁矿表面Acidithiobacillus ferrooxidans菌的EPS组分 (2 mol葡萄醛酸络合物+1 mol Fe3+ =1 mol正电荷) , 他们认为带正电的细菌通过静电吸引作用吸附于溶液中带负电的黄铁矿表面
[31 ,32 ]
。Bellenberg等
[33 ]
通过比较添加/未添加三价铁和葡萄糖酸钠体系的微生物吸附效果发现添加组黄铁矿的覆盖率在1天和8天后分别是未添加组的5倍和8倍, 就此推断吸附能力的上升是溶液中添加的三价铁与EPS的螯合作用增加了细菌的正电, 从而促进了吸附。然而Harneit等学者
[34 ]
发现以硫为能源的培养的Acidithiobacillus thiooxidans和Acidithiobacillus ferrooxidans菌的EPS中并没有发现铁离子络合物;Lara等
[35 ]
就此结果认为Acidithiobacillus thiooxidans在黄铁矿上的吸附并不是靠静电作用的吸附。Tan和Chen
[36 ]
使用多聚赖氨酸对石英表面改性, 使石英表面zeta电位由负变为正, 通过比较改性前后Acidithiobacillus ferrooxidans菌起始阶段的吸附动力学发现二者差别不大, 进而得出结论静电作用不是细菌与矿物亲和度的原因所在, 而主要由二者表面的亲水性决定。此外, Bos等学者
[37 ]
对起始阶段细菌与基底的物理化学作用作了广泛的总结, 在此不再展开重复。第三个阶段是稳定吸附阶段。经历不可逆吸附过程的细菌表面化学组成发生变化。González等
[38 ]
使用激光共聚焦显微技术在研究Acidithiobacillus thiooxidans菌在经过电化学处理过的黄铁矿表面吸附时发现其EPS中的脂类和多糖在15~72 h达到极值, 同时生物层中的单层细胞数目在此时间段内达到最大。Zeng等
[39 ]
发现吸附于矿物表面的细菌增加分泌脂多糖, 表面蛋白或其他胞外聚合物, 而这些物质的分泌有助于生物膜的形成。与此同时, 细菌表面的附属物如菌毛也会进一步加强细菌与矿物表面的作用。总之, 第三阶段细菌增加分泌胞外聚合物与矿物表面形成较强作用力防止细菌脱附。最后, 第四阶段为细菌在矿物表面生物膜内生长繁殖以及吸附新形成的细胞, 此过程大量学者提出了不同的模型, Monds和O'Toole
[40 ]
对微生物定殖以及生物膜发展做了系统的总结, 在此不再展开论述。
Acuna和Meyer等学者
[41 ,42 ]
发现一些Acidithiobacillus ferrooxidans和Leptospirillum ferrooxidans菌种具有对Fe (II) / (III) 浓度梯度和硫代硫酸盐等离子积极趋向的化学感应系统。而到目前为止, 细菌在矿物表面特定位点的感应与吸附还是一个有争论的议题。Becker及其同事
[43 ]
使用原位原子力显微镜成像技术对Sulfobacillus thermosulfidooxidans在黄铁矿表面吸附进行的成像研究并未发现Sulfobacillus thermosulfidooxidans菌种在黄铁矿矿片表面的选择性吸附。Pace等
[44 ]
在研究Acidithiobacillus ferrooxidans菌在黄铁矿表面吸附定殖时也得到类似结论, 他们认为细菌并不优先吸附于黄铁矿表面的缺陷处。而Tan和Chen等
[36 ]
通过对Acidithiobacillus ferrooxidans菌在不同溶液条件 (培养基, p H值) 吸附于多种矿物 (黄铁矿, 黄铜矿, 石英, 斑铜矿, 原混矿) 起始阶段的吸附动力学研究发现细菌覆盖率均小于3%。这表明细菌于矿物表面的吸附是具有高度位点选择性的, 而在适宜生活的环境下可以增加矿物表面的活性位点即增加吸附。Vera等
[25 ]
的一篇综述中介绍了他们的研究, 他们利用kelvin探针联合原子力显微镜研究Leptospirillum ferrooxidans在黄铁矿表面的吸附发现Leptospirillum ferrooxidans与黄铁矿接触区域黄铁矿表面具有更负的电位差 (相对于探针) 。他们认为细菌通过化学感应系统瞬间被吸引于矿物表面特定位点。他们推测这种位点主要位于铁硫原子比不为1∶2的晶格缺陷处;结晶过程中混入其他原子处;和/或结晶过程中温度变化而导致的无定型晶体结构处。此外还有一些研究支持细菌与黄铁矿的吸附不是随机结合而是定位吸附
[28 ,29 ,45 ,46 ,47 ]
。
3 EPS
胞外聚合物 (EPS) 被定义为“生物层中具有使细胞之间以及细菌与其他微粒物质相互内聚或与基质粘附功能的细菌分泌的一种有机聚合物”
[48 ]
。它位于革兰氏阴性细胞的细胞壁或革兰氏阳性细胞外膜之外。Teschke
[49 ]
使用电子力显微镜 (AFM) 对Acidithiobacillus ferrooxidans菌的研究发现EPS的厚度在20~600 nm之间, EPS中所含单个细胞处的厚度为400~100 nm。
3.1组成与提取方法
EPS的物质组成及含量具有高度可变性, 即使同一菌种在不同条件下的组成都不尽相同。它主要由生物大分子组成, 其中主要包括蛋白质, 多糖, 脂, 核酸, 及其复合物 (多糖蛋白质复合物) 。对于Acidithiobacillus ferrooxidans R1菌种在黄铁矿表面形成的EPS, Gehrke等
[28 ,29 ]
的研究表明它的组分包括糖类 (葡萄糖, 鼠李糖, 木糖, 海藻糖, 甘露糖) , C12-20饱和脂肪酸, 葡萄糖醛酸和三价铁离子。EPS的组成决定了它的功能, 对于EPS的提取可以分为化学和物理两大类方法, 表1
[50 ]
提供了一些EPS的提取分离方法可供相关研究人员参考。
3.2影响EPS的因素
影响EPS产生的数量、组成、厚度的因素很多例如菌种, 培养/吸附基质, 浸出液组成及浓度, 溶液p H以及接种时间等。Xia等
[59 ]
通过对Acidithiobacillus ferrooxidans ATCC菌种在单质硫, 黄铜矿以及石英砂上吸附的研究表明不同基质致使细菌产生的EPS中多糖和蛋白质的比例不同, 而这也正是细菌的吸附速率不同的原因。Zeng等学者
[39 ]
通过对黄铜矿浸出过程中, 中温混菌EPS成分的数量及其组成的分析得出结论:起初EPS的数量增长很快并达到稳定, 但是醛糖酸和三价铁的比率随着浸出时间而变化。Bellenberg等
[33 ]
通过比较添加铜离子与未加入铜离子Acidithiobacillus ferrooxidans T菌在黄铁矿表面形成EPS的红外光谱图发现其在组成上存在差别。Gehrke等
[28 ]
研究表明以单质硫为能源培养的细菌EPS组成相对于以黄铁矿为能源的细菌EPS中有较少的醛糖酸和糖类, 但是有较多的脂。Bellenberg等
[60 ]
发现添加葡萄糖或半乳糖会有助于夹膜多糖 (CPS) 的生物合成, 从而帮助EPS的生成。
Gonzalez等
[61 ]
发现C-14 AHL和C14-AHL混合物的加入加强了Acidithiobacillus ferrooxidans在黄铁矿和硫表面生物层的形成和EPS的合成。Becker等
[43 ]
使用原位原子力显微镜成像技术对Sulfobacillus thermosulfidooxidans在黄铁矿表面吸附进行研究发现EPS的厚度随溶液p H值呈现应激性变化。EPS的中大分子聚合链的重排是导致微生物适应外界酸性增加而EPS厚度减小甚至脱附的原因。虽然中外学者对EPS已经展开了相当多的研究, 但是对于浸矿微生物根据环境以及基质分泌不同数量和组成的EPS的分子机制还远远没有清楚。

3.3浸矿功能

研究发现EPS中多糖-蛋白质复合物是细菌与基质黏附的主要物质。EPS作为一种胶状粘性物具有黏附基底和保护群落与有毒物质分离的功能。由于Acidithiobacillus ferrooxidans和Leptospirillum ferrooxidans在黄铁矿表面的EPS中三价铁与醛糖酸形成带一个正电的三价铁络合物, 在EPS中细菌外膜与矿物表面之间形成反应区域, 三价铁络合物可以与黄铁矿直接反应从而加快黄铁矿溶解 (图1)

发现Sulfobacillus thermosulfidooxidans通过EPS的不同表达以应激环境中酸度的增加。总之, EPS作为一种不是细菌生存所必须的分泌物
[40 ]
, 主要功能为保护细菌适应环境;而EPS中三价铁络合物因其与溶液中游离的Fe3+ 离子存在物理化学性质方面的差异 (如粘度, 电导率等) , 其是否能与黄铁矿反应, 以及EPS中是否具有微生物的主动的代谢过程和机制还不甚了解, 需要进一步从实验和理论计算等方面予以研究和证实。

4总结
尽管近年来对于浸矿微生物在黄铁矿氧化中的接触作用已经进行相当的研究, 但是对于一些特定问题仍不甚了解, 针对于目前仍存在争议的议题进行总结以供研究者参考。通过对以下不同方面问题的研究, 将有助于更好地优化/抑制生物浸出作用, 从而根据不同的体系进行工业优化以及产酸控制。
1.决定微生物起始阶段吸附的关键因素是矿物与细菌表面的亲水性/疏水性或静电作用。
2.混菌中哪种微生物优先吸附, 哪种需要其他菌种的预先吸附。
3.微生物是否选择性吸附于表面能高的位置, 是否具有特定位点的感应系统, 其选择性吸附的机制是什么。
4.溶液中添加金属离子, 螯合剂, 表面活性物质等对微生物吸附有何种程度的影响。
5.EPS中三价铁络合物的物理化学性质与溶液中游离的Fe3+ 存在差异, 这种络合物能否氧化黄铁矿及其氧化速度快慢需进一步证实与研究。
6.不同学科领域的方法引入研究, 如生命科学中蛋白质组学, 转录组学, 代谢组学等方法对于不同种类浸矿微生物的系统研究。
