艾叶挥发油明胶微球的制备及其性能表征

摘    要:制备新型艾叶挥发油明胶微球,探究其最佳制备工艺,并对其形态学性质和载药性进行研究,以增加艾叶挥发油在制剂中的稳定性,从而增强其药物疗效。实验中以天然的生物可降解明胶为载体,采用乳化交联法用不同复合凝聚剂制备微球,并通过考察微球粒径及分布、载药量和包封率等指标,采用正交试验设计法优化微球的制备工艺;通过显微镜、扫描电镜和紫外分光光度法等对微球的性能进行研究。研究结果表明:在最佳制备工艺条件下,采用复合凝聚剂所制备的艾叶挥发油明胶微球形态圆整,粒径均匀,分散性良好,平均粒径为23.25 μm,平均载药量(质量分数)为8.52%,药物包封率为29.35%,重现性较好;优选制备工艺所得微球具有较强的储存稳定性和操作稳定性,工艺具有可行性。

Abstract: An efficient procedure was selected to prepare a new type of the gelatin microspheres of volatile oils from Folium Artemisiae Argyi, and its morphology property and drug loading were explored to improve the stability of volatile oils from Folium Artemisiae Argyi in medicine. In the experiments, degradable gelatin was used as a carrier, and the microspheres were prepared by emulsifying and cross-linking technique. Orthogonal design method was used to obtain the best technique, with the diameter of the microspheres, its distribution, utilization ratio of oil, and drug loading amount, envelop rate as the indexes. The product was characterized via microscope, scanning election microscope and UV spectrophotometry. The results show that the product prepared according to the optimal technique with composite cohesion agent is in good shape with smooth surface and even in size, with an average diameter of 23.25 μm, an average drug loading rate of 8.52%, envelop rate of 29.35% and good reproducibility. Furthermore, the established preparation technique is practical and enhances storage stability and good durability in the repeated use.
