采用酶法提取荷叶中的荷叶碱

摘    要:以晒干粉碎的荷叶为原料,采用纤维素酶预处理,稀盐酸浸提,超声波辅助提取,氯仿萃取方法提取荷叶碱。考察纤维素酶用量、酶解温度、酶解时间、酶解酸度、液料比、稀盐酸浸提时间、浸提液中盐酸浓度对荷叶碱提取率的影响,经紫外分光光度计检测所得荷叶碱浓度,确定最佳实验条件。研究结果表明,最佳提取工艺条件是:酶的用量为2.667×10-6 mol/(s·g),酶解温度为50 ℃,酶解时间为2 h,酶解酸度pH=5.0,液料比为30:1,浸提时间为16 h,盐酸浓度为0.5%,荷叶碱的提取率达到1.105%。

Abstract: Dried and smashed lotus leaf was used as raw materials. Nuciferine was extracted by pretreatment using cellulase, immersed into diluted hydrochloric acid, extracted by ultrasonic and chloroform, respectively. The influence of enzyme amount, enzymolysis temperature, enzymolysis time, pH of enzyme solution, the ratio of solvent volume to mass of raw materials, extraction time,concentration of hydrochloric on extraction efficiency was investigated. The optimal condition was achieved by determining the concentration of nuciferine through UV. The results show that the optimal condition of extraction is as follows: The cellulase amount is 2.667×10-6 mol/(s.g), enzymolysis temperature is 50 ℃, enzymolysis time is 2 h, enzymolysis acidity pH=5.0, the ratio of solvent volume to mass of raw materials is 30?1, extraction time is 16 h, and concentration of hydrochloric is 0.5%. The extraction ratio of lotus leaf reaches 1.105%.
